چکیده

Lines of evidence suggest that hypothalamic neuropeptides have a physiological role in the regulation of food intake in mammals. However, little is known about the age-dependent change of hypothalamic neuropeptides in chicks. In this study, we investigated the age-dependent changes in hypothalamic mRNA levels of neuropeptide Y (NPY, an orexigenic neuropeptide), proopiomelanocortin (POMC, the precursor of anorexigenic neuropeptides), and corticotropin-releasing factor (CRF, a possible mediator in the POMC-induced anorexigenic pathway) in growing broiler chicks. The weights of body and abdominal adipose tissue of chicks were measured at 7, 14, 21, and 28 days of age, and the hypothalamic mRNA levels of NPY, POMC, and CRF were analyzed by real-time PCR. Hypothalamic NPY mRNA levels at 14 and 28 days of age were significantly (P<0.05) lower than that at 7 days of age. Hypothalamic POMC mRNA level at 28 days of age was significantly lower than that at 7 days of age. Hypothalamic CRF mRNA level was not affected throughout the experimental period. Abdominal adipose tissue weight was significantly increased after 14 days of age in chicks. Our findings demonstrated that the expressions of NPY and POMC are dependent on age in boiler chicken hypothalamus.

Effect of 24 h Fasting on Gene Expression of AMPK, Appetite Regulation Peptides and Lipometabolism Related Factors in the Hypothalamus of Broiler Chicks

The 5'-adenosine monophosphate-activated protein kinase (AMPK) is a key part of a kinase-signaling cascade that acts to maintain energy homeostasis. The objective of this experiment was to investigate the possible effects of fasting and refeeding on the gene expression of hypothalamic AMPK, some appetitive regulating peptides and lipid metabolism related enzymes. Seven-day-old male broiler (Arb...

Peripheral Insulin Doesn’t Alter Appetite of Broiler Chicks

An experiment was conducted to investigate the effect of peripheral insulin treatment on appetite in chicks. Six-d-age chicks with ad libitum feeding or fasting for 3 h before injection received a subcutaneous injection of 0, 1, 3, 5, 10, or 20 IU of insulin or vehicle (saline). The results showed peripheral insulin treatment (1 to 20 IU) did not alter significantly the feed intake in chicks un...

Differential Changes in Expression of Stress- and Metabolic-Related Neuropeptides in the Rat Hypothalamus during Morphine Dependence and Withdrawal

Chronic morphine treatment and naloxone precipitated morphine withdrawal activates stress-related brain circuit and results in significant changes in food intake, body weight gain and energy metabolism. The present study aimed to reveal hypothalamic mechanisms underlying these effects. Adult male rats were made dependent on morphine by subcutaneous implantation of constant release drug pellets....

Effects of acute heat stress on gene expression of brain-gut neuropeptides in broiler chickens.

Heat stress-induced reduction in feed intake is an annoyance of the poultry industry. Feed intake is regulated by complex mechanisms in which brain-gut neuropeptides are involved, but the changes in such neuropeptides in broiler chickens during heat exposure remain unclear. In this study, we investigated the effects of acute heat stress (35°C, 6 h, and 65% relative humidity) on the gene express...

Leptin regulates appetite-related neuropeptides in the hypothalamus of developing rats without affecting food intake.

Leptin regulates food intake in adult mammals by stimulating hypothalamic anorexigenic pathways and inhibiting orexigenic ones. In developing rodents, fat stores are low, yet circulating leptin levels are high and do not appear to regulate food intake. We determined whether two appetite-related neuropeptides [neuropeptide Y (NPY) and proopiomelanocortin (POMC)] and food intake behavior are sens...
